Our research focuses on the application of mass spectrometry to the study of protein complexes. By careful control of conditions we have demonstrated not only that protein complexes can be maintained intact but that their dynamic interactions and compositional heterogeneity can be studied as a function of time, cell cycle and modification. Recently we have extended our approaches to intact membrane protein complexes, defining their overall subunit stoichiometry and investigating the effects of small molecule binding on their overall conformation.
