Our main goal is to unravel the nature of DNA replication origins in multicellular eukaryotes. We identified an Origin G-rich Repeated Element (OGRE) that could form G quadruplexes, orientated 150-300 bp upstream of the initiation site in a nucleosome-free region. We also demonstrated its functional importance. We also revealed a specific organisation of origins along TAD domains, which is disorganised in cancer cells.
