I use bulk solution and single filament kinetics and thermodynamic analysis of actin filament assembly in vitro to understand the molecular mechanisms by which polarized actin assembly is regulated in motility. I characterize the function of all main regulators of actin assembly. My goal is to challenge the derived mechanistic insight to reconstitute motile processes in vitro with a minimal number of pure components.
