We study RNA polymerase arrest at lesions and in unusual DNA sequences, to learn mechanisms of transcription-coupled DNA repair (TCR). Cockayne syndrome (CS) and UV-sensitive syndrome are TCR deficient and present no cancers, but CS cells are sensitive to reactive oxygen species. We have developed an ultrasensitive comet-FISH assay to measure oxidative damage and its repair in expressed genes.
