The Ubiquitin Proteasome System is a key element of cellular homeostasis. Central to this process are ubiquitin ligases that modify target proteins with ubiquitin. We analyse their function using biochemical methods to understand the underlying molecular principles. In particular, we focus on protein homeostasis and the involved ubiquitin ligases in the endoplasmic reticulum (ER) in a process termed ER associated protein degradation (ERAD).
