Aneuploidy and polyploidy are major deviations from normal chromosome number. They act as key drivers of cancer evolution. Despite their prevalence in tumors, their functional consequences remain poorly understood. We develop models of aneuploidy and polyploidy to systematically dissect their genomic, transcriptomic, and phenotypic effects, and to determine how they promote cancer initiation, progression, and therapy resistance.
